基于低场核磁共振和物性分析技术的牡丹皮饮片干燥特性及动力学研究

目的: 量化表征牡丹皮饮片干燥过程的水分、物性变化,为阐明牡丹皮饮片干燥机制、优化干燥工艺提供科学依据。方法: 构建牡丹皮饮片干燥特性曲线,研究其干燥动力学特征;采用低场核磁共振及其成像技术(LF-NMR/MRI)和物性分析技术,分别研究牡丹皮干燥过程中的水分相态、分布迁移变化和物性特性变化;采用HPLC法考察牡丹皮饮片干燥过程中指标成分丹皮酚和芍药苷的质量分数变化。结果: 55 ℃干燥时,牡丹皮饮片具有恒速干燥阶段和降速干燥阶段,需干燥5 h。LF-NMR/MRI结果显示,干燥过程中,水分由外向内散失;水分相态发生显著变化,结合水比例先大幅度增长后小幅度下降;干燥2 h后,检测不到牡丹皮水分成像信息。各物性指标随着干燥时间的延长而变大,前2 h内随干燥时间变化不明显,2 h后变化显著。干燥5 h内,牡丹皮饮片芍药苷、丹皮酚质量分数无明显变化。结论: 干燥动力学、低场核磁共振和物性分析技术可直观量化表征牡丹皮饮片干燥过程,为阐明牡丹皮饮片干燥机制、优化干燥工艺提供理论依据。

Objective: By quantitatively characterizing the changes of moisture and physical properties, the research aims to provide scientific basis for elucidating the drying mechanism and optimizing the drying process of Moutan Cortex decoction pieces. Methods: The drying characteristic curve was established to study the drying kinetic characteristics of Moutan Cortex decoction pieces. The water phase, distribution, migration and physical properties of Moutan Cortex were studied by LF-NMR/MRI and physical properties analysis technology during drying. Furthermore, HPLC was used to investigate the content changes of paeonol and paeoniflorin which are the indicative components of Moutan Cortex decoction pieces during the drying process. Results: When dried at 55℃, Moutan Cortex decoction pieces had a constant-rate drying period and a falling-rate drying period, which needed to dry for 5 hours. The results of LF-NMR/MRI showed that the water dissipated from outside to inside during the drying process. The water phase changed significantly, and the proportion of bound water increased greatly first and then decreased slightly. After drying for 2 hours, water imaging information of Moutan Cortex could not be detected. With the extension of drying time, all physical properties were increased, and the changes were not obvious within the first 2 hours, but were significant after 2 hours. Within 5 hours of drying, the content of paeoniflorin and paeonol in Moutan Cortex decoction pieces did not change significantly. Conclusion: The combination, including drying kinetics, LF-NMR/MRI and texture properties analysis techniques, can characterize the drying process of Moutan Cortex decoction pieces directly and quantitatively, which provides the theoretical basis for illustrating and optimizing the drying process.
