采用水提醇沉法提取纯化芦荟多糖,复配玉米低聚肽,测定其体外自由基清除能力,并且通过酒精性肝损伤小鼠模型验证其保肝作用。体外实验表明,芦荟多糖复方对DPPH、ABTS和超氧阴离子的清除率比单一成分有较大的提高。动物实验表明,与空白组相比,模型组小鼠血中ALT和AST活性明显升高(P<0.05);抗氧化酶GSH、SOD、GSH-Px活性显著性降低(P<0.01),而丙二醛含量明显增加,炎症因子TNF-α以及IL-6水平明显增高;病理切片显示模型组小鼠肝细胞明显的炎性浸润和空泡变性。芦荟多糖联合玉米低聚肽复方可降低ALT、AST、TC、TG的水平至(25.27±0.90)U/L、(122.50±4.28)U/L、(2.82±0.20)mmol/L、(2.00±0.15)mmol/L,降低MDA含量至(1.29±0.23)pmol/mg,提高GSH、SOD、GSH-Px含量至(95.85±3.93)、(62.04±3.03) U/mg和(80.15±6.04)U/mg;降低IL-6和TNF-α水平至(28.91±0.35)、(157.88±8.67)pg/mg (P<0.05),表明芦荟多糖复方具有不同程度的保肝效果,其主要机制是清除体内体外自由基、提高肝脏抗氧化能力,缓解酒精引起的氧化损伤和炎症。
The aloe polysaccharides were extracted and purified by water extraction and alcohol precipitation,and the oligopeptides of maize were compounded to determine the free radical scavenging ability in vitro.The liver protection effect was verified by a mouse model of alcoholic liver injury.In vitro experiments showed that the aloe polysaccharide compound had a higher clearance rate for DPPH,ABTS and superoxide anion than single component.Animal experiments showed that compared with the blank group,the blood levels of ALT and AST in the model group were significantly increased (P<0.05);the activities of antioxidant enzymes GSH,SOD and GSH-Px were significantly decreased (P<0.01).The content of malondialdehyde increased significantly,and the levels of inflammatory factors TNF-α and IL-6 were significantly increased.Pathological sections showed obvious inflammatory infiltration and vacuolar degeneration of mouse liver cells in the model group.Aloe polysaccharide combined with corn oligopeptide complex can reduce the levels of ALT,AST,TC and TG to (25.27±0.90) U/L,(122.50±4.28) U/L,(2.82±0.20)mmol/L,(2.00±0.15)mmol/L,reduce MDA content to (1.29±0.23) pmol/mg,increase GSH,SOD,GSH-Px content to (95.85±3.93),(62.04±3.03) and (80.15±6.04)U/mg;decreased IL-6 and TNF-α levels to (28.91±0.35),(157.88±8.67)pg/mg (P<0.05),indicating that aloe polysaccharide compound has different degrees of liver protection,the main mechanism is to clear free radicals in vitro and in vivo,improve liver antioxidant capacity,and alleviate oxidative damage and inflammation caused by alcohol.
