响应面分析法优化蛹虫草菌糠多糖提取工艺的研究

以蛹虫草(Cordyceps militaris)菌糠为材料,进行菌糠多糖提取条件的优化研究。单因素实验表明,提取温度、水料比及提取时间均不同程度地影响多糖提取率。根据Box-Behnken中心组合实验设计原理对提取温度、水料比、提取时间进行三因素三水平试验设计,采用响应面软件Design-Expert进行处理,获得蛹虫草菌糠多糖提取最佳的优化条件。即提取温度100 ℃,水料比50∶1,提取时间3 h 优化条件下多糖提取率达5.45 %, 多糖提取预测值为5.88 mg·mL-1。建立的模型拟合度较好,可用来对蛹虫草多糖提取工艺的分析和预测。

Single-factor experiments and Box-Behnken central composite design were used to optimize the extraction of polysaccharides from waste materials of Cordyceps militaris. The single-factor experiment results showed that three factors including the extraction time, water-materials ratio, and extraction temperature of the experiment would affect the extraction degree. According to Box-Behnken central composite design principle, three factors and three levels experiments are designed, and optimal conditions of extraction of polysaccharides are obtained. Response surface processing software Design-Expert was used. Results showed that optimal extraction conditions were temperature of 100 ℃, time of 3 h, water-material ratio of 50∶1. The extraction rate of polysaccharide was 5.45 % and predictive value of polysaccharide was 5.88 mg/mL. The model fit well, which can be used to analyze and predict the extraction process of polysaccharide from Cordyceps militaris.
