大黄虫丸对家兔动脉血栓形成模型的影响

目的:观察大黄虫丸对家兔动脉血栓形成模型的作用。方法:家兔65只随机分为正常组、改良挂线模型组、阿司匹林组、氯吡格雷组、阿司匹林加氯吡格雷组、大黄虫丸低剂量组、大黄虫丸高剂量组共7组,灌胃相应的药液8 d,于末次灌胃2 h后处死家兔,取动脉血管。光学显微镜下观察各组家兔颈动脉血管组织?plusmn;硌榭觥2捎妹噶庖呶绞匝椋╡nzyme-linked immunosorbent assay, ELISA)法检测血浆中1,6-二磷酸果糖二钠盐(D-fructose-1,6-diphosphate trisodium salt octahydrate, FDP)、D-二聚体(D-dimer)、组织因子(tissue factor, TF)等含量的变化。结果:改良挂线模型组血管内充满血栓,血管内弹力膜损伤严重;大黄虫丸低剂量组管腔内丝线周围有血栓形成,但有部分软化、溶解、吸收;大黄虫丸高剂量组管腔内血栓较少,大部分已被吸收,血管内膜清晰完好。大黄虫丸低剂量组与改良挂线模型组比较,血栓干湿重及D-Dimer和FDP含量均有明显差异(P<0.01)。大黄虫丸低剂量组与阿司匹林组比较,除血栓湿重外(P<0.01),血栓干重及D-Dimer和FDP含量均无统计学差异(P>0.05)。大黄虫丸低剂量组与氯吡格雷组比较,除FDP含量(P<0.05)外,血栓干湿重和D-Dimer均无明显差异(P>0.05)。大黄虫丸低剂量组与阿司匹林加氯吡格雷组比较,血栓干湿重及D-Dimer和FDP含量均有明显差异(P<0.01或P<0.05)。大黄虫丸高剂量组分别与改良挂线模型组、大黄虫丸低剂量组、阿司匹林组比较,血栓干湿重及D-Dimer和FDP含量均有明显差异(P<0.01或P<0.05)。大黄虫丸高剂量组与氯吡格雷组比较,除D-Dimer外(P>0.05),血栓干湿重及FDP含量均有明显差异(P<0.01或P<0.05);与阿司匹林加氯吡格雷组比较,血栓干湿重及D-Dimer和FDP含量均无明显差异(P>0.05)。结论:大黄虫丸能降低血浆TF和D-Dimer及FDP的含量,抗动脉血栓形成,维持血管内膜的稳定,其疗效与剂量成正比,大黄虫丸是理想的抗血栓药物。

ObjectiveTo investigate the effect of Dahuang Zhechong pills (DZ) on arterial thrombotic model in vivo.MethodsSixty-five rabbits were randomly divided into 7 groups: normal, model (collagen encapsulated thread-drawing),model+aspirin (ASA), model+clopidogrel (CP),model+ASA+CP, model+ low dosage DZ (DZL), and model+high dosage DZ (DZH).All rabbits except the normal group were fed with the drugs repectively for 8 days,and sacrificed at 2 hours after the last feeding, obtained aortae. The pathological changes in the aortae were observed under microscope,and the level of FDP, D-dimer and tissue factor (TF) were measured by enzyme-linked immunosorbent assay (ELISA). ResultsThe vascular vessels were filled with thrombi in the model group and the elastic membranes of the vessel wall were seriously injured. The arterial thrombi were observed around the vascular wall in the DZL group, but some of the thrombi were dissolved. The number of thrombi was remarkably decreased in the DZH group, and most thrombi were dissolved and the vascular intimal membranes were intact. Compared with the model group, the dry and wet weight of the thrombi and the level of D-dimer, FDP, and TF in the plasma were significantly atte-nuated(P&;lt;0.01) in all the treatment groups. There were no significant difference between the DZL group and the ASA group in the dry weight, D-Dimer, and FDP (P&;gt;0.05). The pathological changes in the vascular vessel and the elevation of plasma parameters in the DZL group were similar to those in the ASA and CP groups (P&;gt;0.05). The dry and wet weight, D-dimer, FDP, and TF in the plasma in the DZH group were significantly lower than those in the DZL group (P&;lt;0.01 or P&;lt;0.05, separatively), and closed to those in the ASA+CP group. ConclusionDahuang Zhechong pills are potential novel anti-thromobotic agent for arterial thrombosis.
