黄连解毒汤由黄连、黄芩、黄柏、栀子四味中药组成,主要包含生物碱类(黄连、黄柏),黄酮类(黄芩)和环烯醚萜类(栀子)三大类成分,其理化性质差别较大,在分别进行高效液相色谱(HPLC)分析时适用的流动相差别较大。本文比较了不同浓度的甲酸-三乙胺流动相添加剂对黄连解毒汤水提取物高效液相色谱(HPLC)分析的影响,提出了一种稳定、重复的HPLC分析方法:柱子为HypersilODS2色谱柱(250×4.6mmi.d,5μm),流动相组成为0.3%(V/V)甲酸-0.3%(V/V)三乙胺的水溶液与0.3%(V/V)甲酸-0.3%(V/V)三乙胺的乙腈溶液,检测波长260nm。这种方法能够同时分析复方中的生物碱类、黄酮类和环烯醚萜类物质,为类似的成分复杂中药提取物的分析提供了参考。
The decoction Huanglianjiedutang consists of four medicinal plants, i.e. Coptis japonica Makino, Scutellaria baricaleins Georgi, Phellodendron amuranse Ruprecht and Gardenia jasminonoides Ellis and its constituents mainly include three groups of flavonoids, alkaloid and iridoid glycosides with great difference of chemico-physical properties, which assume different mobile phases when analyzed by HPLC respectively. This article describes via comparison the influence of formi acid-triethylamine mobile phase additive with different concentrations on the analysis of the aqueous extract of the decoction Huanglianjiedutang by HPLC, thereby having developed a stable and repetitive method of HPLC analysis, in which the analysis is composed of a chromatographic column Hypersil ODS2(250 × 4.6mm i.d, 5 um particle size), a mobile phase consisting of the aqueous solution of 0.3%(v/v) formic acid-0.3%(v/v) triethylamine and the acetonitrile solution of 0.3%(v/v) formic acid-0.3(v/v) triethylamine, and an ultraviolet detector with the wavelength 260nm. This method can be used to analyze alkaloids, flavonoids and iridoid glycosides of TCM compound drugs at the same time, thas providing a refernce for the analysis of TCM extracts with complex constituents.
