通过噻唑蓝(MTT)比色法检测了两种N,N-二烷基脲化合物和四种烷基酚聚氧乙烯醚系列表面活性剂对人宫颈癌细胞(HeLa) 及中华仓鼠卵巢细胞(CHO)的细胞毒性,探讨两类物质对细胞毒性的大小。结果表明: N,N-二烷基脲及烷基酚聚氧乙烯醚对细胞的增殖抑制率均随其浓度的增大而增大。 N,N-二乙基脲和N,N-二丙基脲浓度达到10mg/ml时,对两种细胞有较强的抑制增殖作用,抑制率均大于80%。烷基酚聚氧乙烯醚OP-7、OP-9、NP-10和NP-25的浓度分别达到0.1mg/ml、0.1mg/ml、0.1mg/ml和0.5mg/ml时,对细胞有较强的抑制增殖作用,抑制率均大于80%。因此可知,这六种物质达到一定浓度后,对HeLa细胞及CHO细胞的生长有明显的抑制作用,并呈现浓度依赖性。N,N-二烷基脲对两种细胞的毒性远小于烷基酚聚氧乙烯醚。
MTT assay was adopted to study the proliferation inhibiting effect of two kinds of N,N-dialkyl urea compounds and four kinds of alkylphenol ethoxylates surfactants on HeLa cells and CHO cells,and the cytotoxicity of the two kinds of non-ionic surfactants were discussed.The results indicated that the proliferation inhibiting effect of N,N-dialkyl urea and alkylphenol ethoxylates on both HeLa cells and CHO cells gradually increased as the concentration increased.N,N-diethyl urea and N,N-dipropyl urea could inhibit the proliferation of HeLa cells and CHO cells at the concentration of 10 mg/mL,and the inhibition rate was more than 80%.Alkylphenol ethoxylates OP-7,OP-9,NP-10 and NP-25 could inhibit the proliferation of HeLa cells and CHO cells at the concentration of 0.1,0.1,0.1,0.5 mg/mL,and the inhibition rate was more than 80%.Therefore,when the six substances reach a certain concentration,they inhibits the proliferation of HeLa cells and CHO cells significantly,and the cytotoxicity of N,N-dialkyl urea compounds is much lower than that of alkylphenol ethoxylates.
