目的 建立测定双花解毒合剂中绿原酸含量的高效液相色谱法.方法 色谱柱为Hypersil C1s柱(250 mm×4.6mm,5μm),流动相为0.2 mol/L磷酸二氢钠溶液(用磷酸调节pH至3.2)-甲醇(87∶13),检测波长为327 nm,流速为1.0 mL/min,柱温为40℃.结果 绿原酸质量浓度在36.4 ~ 182.0 μg/mL范围内与峰面积积分值呈良好线性关系(r=0.999 1,n=5),平均回收率为98.92%,RSD为0.83%(n=6).结论 该方法简便快速、准确、重复性好,可用于该制剂的质量控制.
Objective To establish a method for determining the content of chlorogenic acid in Shuanghuajiedu Mixture by high performance liquid chromatography(HPLC). Methods The analysis was carried out at 40 ~C on the Hypersil C ls column(250 mm ×4.6 mm, 5μm). The mobile phase consisted of methanol-0.2 mol/L sodium dihydrogen phosphate solution(13 : 87). The flow rate was 1.0 fnL/min. The detection wavelength was 327 nm. Results The calibration curve of chlorogenic acid was linear over the range of 36.4 - 182.0 μg/mL (r =0. 999 1, n = 5). The average recovery rate was 98.92%, RSD =0.83% (n =6). Conclusion This method is simple, rapid and accurate with good reproducibility and can be used for the quality control of this preparation.
