从基因水平建立名贵春兰品种的鉴定方法,突破以往只能用传统方法鉴定名贵春兰品种的局限性;利用AFLP分子标记技术对18种主要名贵春兰品种进行了DNA指纹图谱分析,结果筛选出16对引物,这16对引物在18个名贵春兰品种中共扩增出345条带(平均每对引物扩增21.56条带),其中有多态性带169条(平均每对引物扩增多态性带10.56条),多态性比例为48.99%;通过这些多态性条带的不同组合,可明确将供试的18个名贵春兰品种加以区分,并作为18个名贵春兰品种鉴定的依据。
To set up a new identification and analysis method of Cymbidium goeringii(Rchb.f)Rchb.f based on the level of gene has broken through the restriction of the traditional method. AFLP fingerprinting analysis on 18 rare varieties of Cymbidium goeringii(Rchb.f)Rchb.f was carried out .16 primer-pairs were screened out and amplified 345 nucleotide acid fragments in the 18 rare varieties of Cymbidium goeringii(Rchb.f)Rchb.f, which the polymorphism rate was about 48.99 %, the average bands amplified by each primer-pair were 21.6. The different combination of polymorphic bands can distinguish the 18 rare Cymbidium varieties, and can be used to identify 18 rare varieties of Cymbidium goeringii(Rchb.f)Rchb.f.
